If you're listing through a real estate agent, he/she should put it into MLS and most (not all) MLS systems will syndicate with Zillow within 24-72 hrs.
If you are doing FSBO (For Sale by Owner - without enlisting an agent):
Go to vflyer.com and get a free account with them. Create a "house for sale listing" with all of your pictures and description as you did on the MLS. You're allowed up to two listings with the free account so you should be ok.Once you click "Publish", within 48 hrs - your listing will (should) syndicate with Zillow, Yahoo, Trulia, Hotpads, etc.

If you wish to list your home on Zillow as a FSBO, simply claim your home (under "edit" on the property details page), and then on the same page you'll see a red icon that says "list for sale/rent". It is $39.95 for 180 d.
