Any bidding on a new home or a renovation needs to be done on a actual cost base, these estimators sites are worthless .
You need your plans looked at by a Contractor in your area, maybe a few of them and they will work with you defining your details and give you an actual cost.
You need to define your finishes, your kitchen your bath layouts ...and the list goes on.
Get an itemized estimation so that you know exactly what you are paying for, another thread on here is some people that had a contract with a builder that states 'hardwood' now the guy is saying 'pergo' !!! Really...

With a house of that size your 'ball park' could be 100K off and you wouldn't even see it ...and that doesn't help you any.

Try using the cost calculator on this site. While it is still an "estimate", it takes into account things like complexity, size, finish, and locale. It also provides an itemized breakout of estimated costs.

One word of caution...

Building cost estimate is very precise (i.e., it breaks out the costs and produces an impressive itemized list, useful in assessing subcontractor bids). But, do not make the mistake of believing that "precise" is a synonym for "accurate". You local market (and personal connections within the construction industry?) will determine actual costs.

You question is not complete enough.  Is there going to be a basement, is it a true 2 story with the master upstairs, 2 or 3 car garage, is the price of the lot additional?
