First, I would find out if the builder has finished out the neighborhood or are they still building. If you purchase a new home construction in the first phase of a neighborhood, plan on staying for a while. Some areas take years to complete and if you go to sell after a year or two, you will be competing with the new home builder. It's hard to win that battle.
I would also recommend not putting in too many upgrades with the builder unless you enjoy paying premium prices. Instead opt for upgrades and features that would be difficult to do after the sale (for example, a separate tub and shower in the master bath - easier to have the builder do it than trying to do it yourself later).
I also firmly believe you need a real estate agent representing you and your interests if you do plan on purchasing a new construction home. Remember that the on-site salesperson at new home communities work for the builders, not you.
